Does the 2001 Kawasaki Ninja ZX6R take 2 cycle oil?

It's a 4 stroke.. Fuel in tank..
Oil 4-cycle in lower right case.. 10w-40

As long as there is proper four stroke oil in the crank case a bit of 2 smoke oil in the fuel tank wouldn't hurt anything really. Wouldn't help anything either though.

10w-40 (or as the manual states) motorcycle specific oil only.

If you want that unmistakable aromatic 2 stroke oil burn smell buy one of these: 2 Stroke Smoke Candle
